The taxonomic status of the Portuguese oyster Crassostrea angulata and the Pacific oyster Crassostrea gigas has often been a matter of controversy. Based on larval shell morphology, experimental hybridisation and electrophoretic studies of enzyme polymorphism several authors have considered these two species as being synonymous. During the recent years, several genetic studies based on mitochondrial DNA and microsatellites data have provided evidences that the two taxa are genetically distinct although closely related. Highly polymorphic genetic markers can be useful tools for the analysis of parental contributions. The present paper reports 3 experiments using microsatellite markers to analyse parental contributions in controlled in vitro factorial crosses of the cupped oyster Crassostrea gigas. Parentage analysis was eased by the large polymorphism observed at the 3 loci studied. In a cross between 20 males and 20 females, a quarter of the progeny could be assigned to a given parent using a single microsatellite locus showing 28 alleles. In 3 crosses between 5 males and females, all the progeny could be assigned to a family by using 3 loci. During the last five years, a great effort has been made to develop genomic tools in oyster species all over the world. In Europe, the Pacific cupped oyster, Crassostrea gigas, and the European flat oyster, Ostrea edulis received a special attention as they are the key species for local aquaculture. Three species of man.-rove oysters, Crassostrea rhizophorae, C. brasiliana, and C. gasar, have been described along the Atlantic shores of South America and Africa. Because the distribution of these molluscs is of great biological and commercial interest, their taxonomy and distribution deserve further clarification. Therefore, 15 populations were sampled from both continents. Their 16S mitochondrial polymorphism was studied by sequencing and PCR-RFLP analysis. Two haplotypes were identified. Selective breeding strategies require pedigree information over generations, but many species produced in aquaculture are too small to be physically tagged at early stages. Consequently, maintaining a sufficient number of separate families is often needed but costly and logistically difficult. Alternatively, parentage assignment can be obtained using DNA markers. Background: Triploidy can occur in many animal species but is often lethal. Among invertebrates, amphibians and fishes, triploids are viable although often sterile or infertile. Most triploids of the Pacific oyster Crassostrea gigas are almost sterile (named "3n beta'') yet a low but significant proportion show an advanced gametogenesis (named "3n alpha''). Source/description : Genomic DNA was extracted from a whole Pacific oyster (C. gigas) after grinding in liquid nitrogen. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). In order to document further the phenomena of variance in reproductive success in natural populations of the European flat oyster Ostrea Midis, two complementary studies based on natural and experimental populations were conducted. The first part of this work was focused on paternity analyses using a set of four microsatellite markers for larvae collected from 13 brooding females sampled in Quiberon Bay (Brittany, France).
